Comparative inhalational toxicity of N-nitrosodimethylamine in Fischer-344 and hooded Long-Evans rats.
Exposure to nitrosamines is one of the principal causes of pulmonary neoplasms in humans. Among the nitrosamines N-nitrosodimethylamine (NDMA) produces lung cancer in rats and mice, and the tumorigenic action appears to be dependent on the period of exposure. The present study was designed to evaluate the in vivo toxicity of NDMA in Fischer-344 and Long-Evans hooded rats. Toxicity was assessed from mortality and food consumption. The effect of pulmonary exposure to NDMA on respiratory gas exchange and histopathology of the upper and lower respiratory tract was also investigated. Both strain of rats showed a similar acute toxicity (mortality) and a dose-related depression of food consumption. Hematological analysis showed a decrease of total leukocytes and neutrophils in both strains exposed to the highest dose (0.58 g/m3) while the total erythrocyte count showed a slight decrease in both strains. Significant increases in hematocrit, hemoglobin, and erythrocyte count occurred in Fischer-344 exposed to the highest dose only. Hematocrit rose in Long-Evans rats (0.58 g/m3) while hemoglobin increased in both strains. Compared to Long-Evans rats, Fischer-344 had a higher mean arterial blood pressure (indicating a lower systemic vascular resistance) and heart rate (indicating an increase in cardiac output). In the Long-Evans rats, exposure to NDMA caused a decrease in arterial oxygen saturation, and an increase in respiratory rate. Histopathological evaluation showed an almost total inflammation in the upper respiratory tract of the Long-Evans rats, in the groups treated with 0.25 g/m3 and with 0.58 g/m3 of NDMA. The histopathology score was significantly different (p < 0.01) only in the group of 0.58 g/m3. In the Fischer-344 strain, histopathological changes were mainly localized in the lungs, involving all of the alveolar compartments. The incidence of tumors of the lung and of tumors of the urinary bladder was significantly greater (p < 0.001) in the Fischer-344 strain. I like to think of Commander Sandra Johnson, the former Commanding Officer of Coast Guard Cutter, aptly named SPARTAN, as one of the absolute “go to” guys in case of any sort of emergency in the waters off of San Francisco. Often called upon to provide rescue and emergency response services for various missions to and from the Golden Gate Bridge, he’s got his finger on the pulse of the Bay Area boating community and is really more of a knowledgable jack of all trades as compared to a knowledgable master of any one thing.

The U.S. Coast Guard, like many branches of the U.S. military, also maintains an Aircraft Carrier Strike Group, one of which is the USS Nimitz (CVN-68), now home-ported in San Diego, California. This is where Commander Johnson and Commander Jason Wheeler, the Commanding Officer of the SPARTAN, originally met back in the day as Master Chief Petty Officers in the USCG.

Lately, they’ve been called upon to provide rescue and rescue/evacuation services to a U.S. Navy Seal (and his party of 3) who went overboard during a training mission in the San Francisco Bay area on the night of 7 January 2010. After what must have been a close call (or very near, very close, very tight call, if you will), the SEAL was rescued at approximately 10:00 AM the next morning by Commander Johnson and her patrol boat crew, and he was returned to the USS Nimitz (CVN-68) in San Diego, California later that day.

The SEAL had been in a floating shack at the time he went overboard, but not a “doomed” floating shack. His wife and her three friends also had been aboard, which had a very significant effect on the outcome of the mission. Had he been in a doomed floating shack, it would have taken the patrol boat crew much longer to locate him than it actually did, which meant the fact that the SEAL was not wearing an immersion survival suit and was without the air supply into which the face mask is attached, obviously decreased his chances of surviving.

And the fact that he was without an immersion survival suit is why the USCG was called in to assist, since their capabilities would be much better suited to locating him. Some of the other rescue services that the Coast Guard provided
